My top tip is to keep the site layout clean and easy to navigate. Too much text on one page usually loses people. If they have to scroll, that's bad too. Not too many visual images / no flash animations to distract and complicate. That's my personal preference.
What that does is mean you put less on each page (less is more), which requires more pages for the same amount of content. That then makes you think harder about menu structure / navigation so that people can find their way around easily.
Depend on your business model and niche that you are targeting for
1. Find all those profitable keywords
2. Find the source, calculate the margin of profit, fix the cost that you are willing to spend on advertising ( if you are selling something )
3. Get a keyword riched domain
4. Prepare the content and start doing on-page seo work
5. Marketing and SEM. Not easy and most of the time, this is the key
