Tables were not meant for layout, but are simpler to get reliable results with unless you're very familiar with CSS. Divs have many advantages, but are harder to get the "hang" of. Be prepared to delve into many workarounds and tricks to get divs to cooperate properly.

My favorite tool is the html5 boilerplate in combination with 960.gs. Paul Irish is the man. He works on the core jQuery team, Core google Chrome team and he was a creator of the modernizer script for html5.

I'd personally prefer divs.
I believe if you get too crazy with your pages and have too many tables than the page won't load as quickly as the same layout made with Div tags. Page load time is definitely important! Plus, I think DIV tags looks cleaner than tables when looking at source code.
I would say work with tables if you aren't comfortable with div tags yet, but move over to the div tags sooner than later. You won't regret it!
