Can we block SMTP port for few users on the domain.

If they have dedicated IP per workstation/client, then you will be able to filter and block users that want to use SMTP but are not authorized (use iptables or ipchains)... I am not quite sure if you can do it with CPANEL directly...

If they are not signing up to send emails, why would you want them to be able to send emails from the server?
