Do you wish to become a Direct Reseller? If yes, you can sign up for their Reseller Account from their website. If you want to get a Sub-Reseller Account, you can try WHMCS

I guess you need to ask them about that or at least check their web site there.

Enom has a setup fee with their Slabs...I would suggest you to check this link: http://www.enom.com/pricing.asp

Enom prices seems to be a little expensive for me.
That's possible to find cheaper IMO.
@ OP, why do you want to have deals only with those two companies?
Why don't you try to find better and cheaper offers?

You can try ResellerClub as the deposit can be used by you. Else, you can sign up for an ENOM/ResellerClub Sub-Reseller Account from WHMCS. WHMCS offers you good pricing to start with

resellerclub resellers do require some initial deposit but there is no yearly service payment like g[x]daddy, so in order to execute your orders you need to make deposit to your account. You could get an account from a reseller of resellerclub so that you directly avail discounted rates on domains

I will recommend signing up with NetEarthOne instead. They use the same platform as ResellerClub and doesn't require a hefty setup fee or initial deposit. You only deposit the amount into the account before making a purchase. :)

Actually if you sign-up with any of the resellerclub sub reseller, most do not need you to make any initial deposit to start selling

resell.biz is a good sub-reseller of ResellerClub. You can start buying domains at a very competitive price with a $5 deposit.

I will suggest to be a reseller instead for being sub reseller. Prefer the registrar mostly used in your area (i mean your competitor uses) because it will be easy to move domain from one cp to other instead of domain transfer process.
